Developed novel process for the regioselective tandem synthesis of variety of polyheterocycles by the copper-catalyzed electrophilic cyclization by the preferential addition of N-Heterocycles on ortho-haloalkynes followed by arylation. Developed novel chemistry allows direct access of various types of diversely substituted N-heterocycles, Carbocycles, Natural products, Synthetic drugs and π-conjugated organic materials. 

Designed a novel cascade reaction for the diversity oriented synthesis (DOS) of over hundred natural-product-likes and π-conjugated scaffolds.

Developed novel chemistry for the Iodine-Catalyzed and Solvent Controlled Selective Electrophilic Cyclization and Oxidative Esterification of ortho-alkynyl Aldehydes: An Easy Access to Pyranoquinolines, Pyranoquinolinones and Isocumarins. We have realized, for the first time iodine-catalyzed selective synthesis of iodo-pyrano[4,3-b]quinolines and Ortho-alkynyl esters from ortho-alkynyl aldehydes.

Novel property of benzotriazole and its derivatives: We had identified the new role of inexpensive and thermally stable compound benzotriazole as an inexpensive and efficient ligand in Copper-Catalyzed C-N (N-arylation), C-S (S-arylation) coupling reaction. 

Development novel and inexpensive ligand for the C-C bond forming Suzuki reaction in water at room temperature (for aryl and heteroaryl iodides and bromides).

Designed novel ligand for the Sonagashira coupling of aryl chlorides with terminal alkynes.

Developed novel method for the Sterioselective synthesis of Quinoxalines: (Reaction in water at room temperature)

Designed a novel method for the synthesis of isoquinolines and naphthyridines by dual activation of electrophile and nucleophile.

Project 1

“Regioselective Tandem Synthesis of Variety of Fused-heterocycles by the Copper and Palladium-Catalyzed preferential addition of N-heterocycles on ortho-haloalkynes followed by C-C bond formation 

This is one of the most exciting and major research projects of our laboratory, which allow direct access of various types of diversely substituted N-heterocycles, Carbocycles, Natural products, Synthetic drugs and π-conjugated organic materials.

In the long history of alkynes electrophilic cyclization chemistry, the synthesis of polyheterocycles by the nucleophilic addition of N-heterocycles onto alkynes, followed by in situ ring closure by C-C bond formation has not much explored. In this project we have successfully synthesized indolo, pyrrolo[2,1-a]isoquinolines and Naphthyridines regioselectively by the copper and palladium-catalyzed preferential addition of N-heterocycles over N-arylation on to alkynes, followed by intramolecular C-2 arylation without isolating enamine intermediate generated in in situ, in the reaction. Application of developed chemistry for the synthesis of other interesting heterocycles and organic materials is currently under way in our laboratory and will be reported in due course.

Project 2

Iodine-Catalyzed and Solvent Controlled Selective Electrophilic Cyclization and Oxidative Esterification of ortho-alkynyl Aldehydes: An Easy Access to Pyranoquinolines, Pyranoquinolinones and Isocumarins

In this project we have developed novel method for the direct synthesis of esters from aldehydes by using iodine as a catalyst. We have realized, for the first time iodine-catalyzed selective synthesis of iodo-pyrano[4,3-b]quinolines 2a-b and Ortho-alkynyl esters 3a-h from ortho-alkynyl aldehydes 1a-j. Developed novel oxidative esterification process, provides a powerful tool for the preparation of wide range functionalized pyranoquinolinones 4a-d as well as isocoumarin 4e.

Chemoselective behaviour of iodine in different solvents in the electrophilic iododocyclization of ortho-alkynyl aldehydes is described. Ortho-alkynyl aldehydes on reaction with I2 in CH2Cl2 with appropriate nucleophiles provides pyrano[4,3-b]quinolines 2, via formation of cyclic iodonium intermediate 5, however using alcohols as a solvent as well as nucleophile, ortho-alkynyl esters 3, were obtained selectively in excellent yield via formation of hypoiodide intermediate 8. Subsequently, ortho-alkynyl esters were converted in to chromones 4, by the electrophilic iodocyclization.

Project 3             

Diversity Oriented Synthesis (DOS) of Over Hundred Natural-Product-Likes and π-Conjugated Scaffolds: A Novel Cascade Reaction.

This is another one of the most challenging ongoing projects of our laboratory, in which we had design a novel cascade reaction for the synthesis of more than hundred scaffolds (achiral and chiral ) in one go. We had successfully established the mechanism for the designed reaction by identifying reaction intermediates by X-Ray crystallographic studies.

We had successfully synthesized and characterized more than twenty scaffolds using this cascade reaction. Synthesis of other designed scaffolds is currently under way in our laboratory.

Project 4

Design of benzotriazole based novel ligands for the copper and palladium-catalyzed coupling (Suzuki, Sonogashira, Heck, Buchwald-Hartwick) reaction

Recently we had identified the new role of inexpensive and thermally stable compound benzotriazole as an inexpensive and efficient ligand in Copper-Catalyzed C-N (N-arylation), C-S (S-arylation) coupling reaction. In this project we had designed several benzotriazole based ligands for the mentioned coupling reactions. Some of the designed ligands efficiently catalyzed the coupling reactions. Work is under progress in our laboratory.
